Objective: To investigate the effects of limited fluid resuscitation (FR) without or with hypertonic saline solution on uncontrolled hemorrhagic shock (UHS) . Methods: A rat Capone's method was used to establish an uncontrolled hemorrhagic shock model that could simulate the life-support chain of emergency medical services for trauma, in the form of 4 distinct phases: Phase I involved 30min of simulated traumatic phase for building UHS model; Phase II involved 60min of simulated first aid phase treated with different fluid resuscitation; Phase III involved 120min of simulated treatment phase treated with hemostasis and blood transfusion; Phase IV involved observing recovery and survival to 72hr. Forty Sprague Dawley rats were randomly divided into 4 groups: 10 into shame-operated group(NC group), which received no model and no FR; 10 into large-volume FR group (NS80 group), which received FR with isotonic saline and MAP was controlled to 80mmHg in Phase II; 10 into limited FR treated with isotonic saline group (NS50 group), which received FR with isotonic saline and MAP was controlled to 50mmHg in Phase II; and another 10 into limited FR treated with hypertonic saline group (HS50 group), which received FR with hypertonic saline and MAP was controlled to 50mmHg in Phase II. NS80 group, NS50 group, and HS50 group received the same procedures after Phase II, Survival rats were followed up to 72hr. The amount of re-bleeding, dosage of infusion in Phase II, and survival rate were observed. Value of PH and BD as well as concentration of MDA, SOD, and TNF- α in serum were detected on a series of time points. All data was analyzed by the SPSS11.0 software.
