| Objective To investigate the effects of Rosiglitazone on the proliferation and ECM synthesis of rat aortic vascular smooth muscle cells (VSMCs) induced by high glucose and the possible mechanism.Methods Vascular smooth muscle cells of Sprague-Dawley rats werecultured and determined by immunohistochemistry. The viability of VSMCs was determined by MTT assay. The cells cycle was examined by flow cytometry. The protein expression of PCNA in VSMCs was evaluated by Western blotting. The mRAN expression of collagen I /III , TGFβ1 of VSMC were determined by RT-PCR. The secretion of TGF β1 in the supermatants was measured by enzyme-linked immunabsorbant assay(ELISA).Result The viability of VSMCs induced by high glucose was increasedand displayed in does-dependent and time-dependent manner. At the concentration of 22Mm D-GS, cells proliferation reached the highest effect at 48 hours ( P<0. 01 ) . Rosiglitazone(10 μmol/l) remarkedly inhibited the viability of VSMCs (P<0. 01), the expression of PCNA (P<0. 05) in VSMCs, decreased the proportion of VSMC "S" periods while increased the proportion of VSMCs G0/G1 periods (P<0. 01) .
