ObjectiveThe vascular stenosis have been caused by the intimal injury and the dramaticly changing of the invasculer hemodynamic, but there are two different mechanisms of stenosis development. We build the two different animal model of vascular stenosis(Carotid Arterial Transluminal Balloon Angioplasty and perivascular common carotid collar placement), and utilize the momordicin on these animals, to investigate the effect of momordicin on vascular stenosis in animal experiment.MethodsAnimal experiment I : using the balloon to catheter carotid arterial of rabbits and reproduce the model of vascular stenosis then fed with 5% high cholesterol mixed forage. 40 purebred New Zealand white rabbits were randomly divided into four groups(control, fake operation ,model and momordicin group). Except the control group, the others three groups were fed with 5% high cholesterol mixed forage. We collected the animal's blood from vein of ear to check the level and changes of TC and NO, ET of the plasma, and got the experiment carotid to make pathology section, dye HE check by pathology morphology. Simultaneously, the image were analyzed with a microscope and HMIAS2000 software to distinguish the rang of intimal and the degree of intravascular stenosis. Then use the immunohistochemistry to analysis the changes of TNF- α, NF- κB, PCNA, P53 protein and immunofluorescence to observe the cell apoptosis of vascular wall.Animal experiment II : The rabbits were operated with perivascular common carotid collar placement to build the animal model of vascular stenosis. 20 purebred New Zealand white rabbits were randomly divided into three groups(control, model and momordicin group).
