With the in-depth research on diabetes mellitus (DM) and its chronic complications, postprandial high glucose PPHG and advanced glycation end products (AGEs) have become hotspot in this field. Clinical researches at home and abroad have paid great attention to blood glucose fluctuation and its damage impact on DM and its complications. The investigation report on blood glucose fluctuation, however, come short, especially about the establishment of fluctuation models and combined damaging impact with AGEs on blood vessel cells. This paper is aiming to explore the establishment of blood glucose fluctuation mouse models, its impact on the mouse organs , the impact of glucose fluctuation on blood vessel cells' morphology and super structure and its impact combined with AGEs on blood vessel cells. Details are as follows:1. Establishment of blood glucose fluctuation mouse models: To explore the damage impact of blood glucose fluctuation on mice through animal model of blood glucose fluctuation. Randomly choose 8 mice from normal and model mice (induced by Alloxan) as normal group, normal fluctuation group,model group and model plus fluctuation group to emulate the blood glucose fluctuation of daily life of ordinary people. Inject glucose into mouse abdomen at 8: 00,12: 00 and 16: 00 respectively. Supervise the whole day blood glucose content every week and detect the glucose tolerance and FDA after 4 weeks. Results show that the establishment of animal models in this way is relatively stable, simple and repeatable. The glucose tolerance mice of model plus fluctuation group is obviously decreased (P<0. 01) , while the FDA level is obviously increase ( P<0. 01 ), indicating that the methodology of establishing blood glucose fluctuation mouse models is feasible.2. Impact of blood glucose fluctuation on mouse organs: Using above animal models to investigate the damaging impact on mice organs after 4 weeks. Results show that the mice's organs of model plus fluctuation group are damaged to different degree, especially the damage of the model plus fluctuation group is more obviously than the model group.
