Lung cancer is the leading cancer-death disease. Gene polymorphic is the major reason for the difference risk of lung cancer among individuals. Everyone may have a unique combination of polymorphic traits that modify genetic susceptibility and response to drugs, chemicals and carcinogens. Genetic Polymorphism is a important regulating type for the gene function, it can influence the protein structure and function by only change a single nucleotide in the coding region.This study investigates the polymorphic of two lung cancer related genes: MDM2 and FUS2. Putative tumor suppressor gene FUS2 encodes a novel cytoplastic N-acetyltransferase located at 3p21.3. Newly identified polymorphism 767A/T leads to a non-conservative amino acid change (R222W) located between the acetyltransferase and the proline-rich domains. This research examines the difference of polymorphism site FUS2-767A/T between the lung cancer cases and the normal controls in Chinese population. PCR-SSCP method was used to determine the genotype FUS2-767A/T in 146 surgically treat patients with lung cancer and 113 normal controls, and test the relationship between lung cancer risk and difference genotypes of FUS2 gene. And find that, FUS2-767A/T shows significant relationship with the cancer type, age, and vessel cancer embolus in clinicopathology (p=0.044, 0.011, 0.031, respectively).Mdm2 is a kind of phosphate protein, can directly bind to p53 protein and inhibit its activity. It amplified in a lot kinds of cancer, and associated with poor response to therapy and prognosis. Recently, a single nucleotide polymorphism (snp309) is found in the promoter and is show to increase the affinity of the transcriptional activator sp1, resulting in higher levels of mdm2 protein and subsequent attenuates the p53 pathway and accelerates tumor formation in humans. We first examine the genotype of MDM2 SNP309 G/T in 151 lung cancer tissue samples with corespongding normal tissues, and then test the expression level of MDM2 and p53 protein within 63 lung cancer tissues.
