Reactor is the key section of bioartificial liver (BAL), and the efficiency of BAL in the treatment of the patients with liver failure counts on the function of the reactor used. Hollow fiber reactor is investigated mostly in the research of BAL owing to shortage of homogeneous hepatocytes, Security hidden danger of liver cell lines and unclear study results of hepatic stem cell. Polysulfone(PS) membrane is fit for mechanical processing and has good thermal stability and resist against oxidation, but its adhesiveness to hepatocytes and influence to the fuctions of hepatocytes are unclear. There are a lot of problems to be resolved about performance of membrane materials, methods of hepatocytes' culture fit for BAL and efficiency of hollow fiber reactor in the research of BAL.In this study, morphology of suckling porcine hepatocytes during primary culture on PS membrane were observed. Synthesis and biotransformation function of the suckling porcine hepatocytes cultured on PS membrane was studied in order to evaluate the applicability of PS membrane as a certain kind of membrane materials of reactor. Magnetism stiring method was applied to form suckling porcine hepatocytes aggregates, morphous and function of aggregates were detected to explore the hepatocytes culture method fit for BAL. PS membrane hollow fiber reactor was applied to construct reactor experimental system, function of suckling porcine hepatocytes aggregates seeded into the reactor's outer space and influence of this system to severe hepatitis plasma(SHP)were detected in order to evaluate the efficiency of PS membrane hollow fiber reactor. The methods and results are following.Changes of the morphology of suckling porcine hepatocytes cultured on PS membrane were observed, synthesis of albumin and urea were detected, clearance of ammonia and enzyme release were examined. All these situations were compared to that of hepatocytes cultured on polystyrene surface. Porcine hepatocytes attached and spreaded on PS membrane surface and formed aggregates. Significant superiority were found in the culture period compared to hepatocytes cultured on polystyrene surface such as synthesis ratio of albumin and urea, clearance rate of ammonia.
