Backgrounds:As an ingredient of combined therapy of cancer, biotherapy can not only treat cancer independently, but also can be combined with surgery, radiotherapy and chemotherapy. It has many advantages,such as higher therapeutic effects, better specificity and minor side effects,which attracts more and more attentions recently. Dendritic cells (DCs) are believed to be the most potential professional antigen presenting cells and has the most powerful antigen presenting capacity. DCs loaded with tumor antigen can act as"tumor vaccine". It can present tumor antigen to immune system, and initiate specific immune response.The presentation of tumor-associated antigen(TAA) by DCs and then recognition by cytotoxic T lymphocytes(CTLs) to induce specific anti-tumor immune response has been a hot spot for tumor therapy.At present most TAAs have tissue specificity for certain tumors,such as MARR-1 specific for melanoma,CEA specific for gastrointestinal or lung cancer.These tumor specific antigens can only induce immune response for the same tumors which express the TAA. In contrast, the CTLs induced by the TAA can not kill other tumors which don't express the TAA. Therefore, it is not only a hot spot, but also a great virgin land to explore in order to look for a universal antigen for various tumors.Ideal tumor universal antigens need to :â‘ be expressed by the vast majority of human cancer, but rarely be expressed in normal tissues.â‘¡be indispensable in the process of tumorigenesis to avoid antigen variation or depletion.â‘¢include peptide sequences that bind to MHC molecules.â‘£be recognized by the T-cell repertoire in an MHC-restricted fashion to elicit specific T-cell response.It is well known that activation of telomerase is an important step for the infinite...
