| Objective: To evaluate the efficacy and safety of topiramate in monotherapy and concomitant therapy for epilepsy .Methods: A non-randomized and controlled clinical study was performed.The patients who have no AEDs enter monotherapy group ,and the patients who were curing by not more than 2 drugs enter concomitant therapy group by strict standard.1.Review time:Medical record of enrollments:seize type , frequency ,head CT or MRI,EEG and other lab data ,drug was noted carefully .We introduce seize observe list to the patients.2.Adding quantity time:8weeks.For adult original dosage of topiramate is 25mg/d in the first week ,increasing 25mg/d every week,and target dose is 200mg/d.For enfant whose avoirdupois during 20-25kg,original dosage of topiramate is 12.5mg/d in the first week ,increasing 12.5mg/d every week,and target dose is 100mg/d;and those whose avoirdupois more than 35kg are same as adult.At the end of the 8th week, we countercheck blood routine test,urine routine test , liver function ,renal function.3. stabilization time:we observe the patients who keeping taget dose for 12 weeks.we visit the patients every month from Review time to the end of stabilization time, and countercheck blood routine test,urine routine test , liver function ,renal function and EEG. The type and dose of AEDs used of the concomitant therapy group is invariable.Results: Both monotherapy and concomitant therapy group show good curative effect and less, unimportance side-effectConclusion: Topiramate in monotherapy and concomitant therapy for seizure is effective , relatively safe and less side-effect. |
