Objective: This research aims to detect the clinical value of PCR combined with DNA sequencing to fungal keratitisLately along with the extensive use of antibiotics and corticosteroid, the incidence of fungal keratitis added distinctly ,and this condition is also associated with diabetes mellitus and the acquired immune deficiency syndrome (AIDS).It have developed an infective diseases of disturbing eyesight and lead to serious result if have no effective and timely treatment. Though clinical history and findings help to the diagnosis of fungal keratitis ,the accurate diagnosis depends on laboratory techniques .Now the diagnosis to fungal keratitis is the potassium hydroxide (KOH) wet mount and culture, that are based on histology and physiology. The potassium hydroxide (KOH) wet mount is simple and quick ,but couldn't recognize the kind of fungiand the culture is credible but too slow.Confocal microscope has the characteristic of rapidness and sensitivity.,but need the cooperation of the patient, what' more, it is so expensive and unfit to extend.Early and accurate diagnosis and reasonable intervention is a critical element for an effective treatmentof fungal keratitis. Since 1995 Alexandrakis et al succeeded in firstly diagnosing Fusarium oxysporum endophthalmitis using PCR, as a quick,sensitive and specific technique.it has become important more and more.
