Background More and more evidence indicates that chronic subclinical inflammation may play important roles in the development of type 2 diabetes mellitus(T2DM) and macroangiopathy. Recently, a new protein which named Tanis had been found in a T2DM and metabolism syndrome animal model. Tanis is a glucose regulated protein with the role of insulin resistance and may be the receptor of serum amyloid A(SAA). The human homologue for Tanis is AD-015/SelS.objective To research the expression of SelS/Tanis mRNA in omental adipose tissue in patients of T2DM and the relationship of SelS mRNA with Homa-IR and serum SAA level. Methods To extract total RNA from human omental adipose tissue. Semi-quantitative RT-PCR was conducted to compare the expression of SelS gene in omental adipose tissue of ten type 2 diabetic subjects with twelve non-diabetic individuals. The serum concentions of total cholesterol, triglycerid, low density lipoprotein cholesterol, high density lipoprotein cholesterol, fasting C-peptide, fasting insulin, fasting plasma glucose and C-response protein were measured at the same time. Calculate insulin resistance index (Homa-IR) and detect SAA level by ELISA. Analyse relationship of SelS mRNA expression in omental adipose tissue with Homa-IR and serum SAA level in all subjects. Results Result of RNA electrophoresis suggests that the total RNA extracted from human omental adipose tissue was...
