Since the development of monoclonal antibodies, it offers a new means to improve the targeting therapy of tumors. Our laboratory has been engaged in targeting therapy study on HCC for more than 20 years. In the past, we have successfully established a monoclonal antibody HA25 against HCC using fresh human HCC cell suspension as antigen, which was proved to have high specificity and affinity by immunohistochemical staining to HCC tissue and radioimmunoimaging. Moreover, the VH and VL gene of monoclonal antibody HAb25 were cloned respectively by PCR, then ligated by linker (Gly4Ser)3 to construct msFv25. The hdsFv with higher stability and lower immuogenicity to human being was obtained by disulfide-stabiliztion and humanized reformation from msFv25 lately. They kept the specificity and affinity of their parent antibody to some extent. In recent years, several kinds of recombinant immunotoxins against HCC, such as scFv25-TNFa,hdsFv25-TNFa mutant, hdsFv25-PE38 and hdsFv25-RC-RNase etcal,have been prepared in our laboratory.
