The developing of tissue engineering has been restricted by the source of seed cells. More and more studies comfirmed that adipose derived stem cells (ADSCs) could be induced into osteoblasts, cartilage cells, nerve cells, myocyte and adipoce cells, etc. Furthermore, adipose tissue can be obtained easily, is easy to autotransplant; and ADSCs could keep multi-directional differentiation when they were cultured in vitro. They had stable genetic background. There were almostly no immunological rejection after transplantation. Because of these advantages, they had been suitable seed cells for tissue engineering.In this research, we studied the biology and plasticity of ADSCs, and investigated whether the combination of ADSCs with PLGA could repair rabbit alveolar bone defects.PART 1Objective: To study the morphology, growth, adhesion, proliferation, cell surface markers, and genetic characters of ADSCs. Methods: ADSCs were isolated from human adipose tissue; the cell morphology was studied by inverted microscope; growth character was determined by cell couting kit-8; cell surface markers and cell cycle were examined by flow cytometry; genetic characters were examined by karyotype analysis. Result: ADSCs showed long fusiform shape; their average population doubling time was about 29h, the cells were positive for CD29, CD90 and CD44; more than 80% cells in exponential phase of growth were in G0/G1; the 20th passage cells still had...
