Background and purposeDendritic cells (DCs) are professional antigen-presenting cells (APC) that can activate naive T cells. Immature DCs can capture and process antigens, while mature DCs present antigens as a result initiating the primary immune response. Many tumor vaccines based on DCs have been developed, but the effects were not so good. So, researchers have focused their eyes on some new vaccines.Exosomes are vesicles secrected by different kinds of cells, such as B lymphocytes, tumor cells as well as mast cells. DCs-derived exosomes (Dexs) express MHC, co-stimulatory and adhesive molecules. Many studies have shown that Dexs can exert anti-tumor immunity in vivo and in vitro. It's a promising cell-free vaccine combating with tumor.CpG ODN is a new adjuvant that can up-regulate the surface molecules, enhance the cytokines production, augment the function of DCs. In this study, we successfully induced DCs from human cord blood , and harvested exosomes from the supernatants of DCs. Three kinds of CpG ODN were used to observe their effects on DCs and Dexs.
