Background and objective:DNA polymerase β (DNA polβ) is a very important repair enzyme in the base excision repair. The function of DNA polβ exerts rightly has a key role in the cell cycle and steady inheritance. DNA polβ is mainly involved in repairing DNA damages resulted by ROS oxidation or deamination of cytosine and met-cytosine. It also may be involved in repairing alkyl bases damage caused by exterior factors such as nitrosamine. Under normal conditions, DNA pol β expresses at a constant low level throughout the cell cycle. However, DNA pol β is in defect of 3'-5' exonuclease activity and the replication fidelity of DNA pol β is very low compared with many other DNA polymerases. Overexpression of DNA polβ could increase genetic instability by interfering other DNA polymerases, furthermore induce the tumor. In our previous studies, mutations of polβgene and overexpression of polβhad been detected in many tumor tissues. p53 gene is the most resprentative antioncogene and wild type p53 protein have a significant effect on the cells growth and inhibiting the cells malignant proliferation. In this study, cell lines stably expressing wild type DNA polβ were established and the cells were suffered with AOH.
