RNA interference (RNAi), as commonly defined, is a phenomenon leading to post-transcriptional gene silencing (PTGS) after endogenous production or artificial introduction into a cell of small interfering double strand RNA with complementary sequences to the targeted gene.In recent years ,the research on RNAi has become the focus. siRNAs are 21-23nt in length . It can especially identify the mRNA transcribled by traget genes and cut some of the areas which are compliment with siRNA antisense therefore inhibit the expression of traget genes. By doing this,RNAi technic will stock some multidrug,and prevent the relevant the expressiong of protein, which has been proved to be valnable.Multidrug resistance (MDR), a major cause of cancer treatment failure, is a phenomenon that cancer cells develop broad resistance to a wide variety of chemotherapeutic drugs. Although the detailed molecular mechanisms of MDR remain to be explored, more people believe that the over-expression of the mdrl result in the increase of the energy-dependent P-glycoprotein is the main reason for the drug resistance. P-glycoprotein may pump out the chemical drug from the cell, so the accumulation of chemical drug decrease. siRNA can knock down the expression of mdrl mRNA and decrease the level of the P-gp for multidrug reversing efficiently. There are already some methods for multidrug reversing to this mechanism.RNAi may be very important to further research on multidrug reversing.we designed this study to investigate the possibility of RNAi on multidrug reversing of gastric cancer cells, two nucleotide sequences of siRNA corresponding to various sites of mdrl gene, were synthesized by transcription in vitro and transfected into gastric cancer cell line SGC-7901/VCR to detect the accumulation of ADM...
