Many studies have showed that expression of B7 gene on tumor cell membrane is essential to cause T cell (major antitumor cell) activation and cytokine generation, because most tumor cells' membrane have not B7 molecules (B7.1 or B7.2), or have less B7 molecules, tumor cells can escape immune identification of body. In addition, in nearly ninety percent of all tumor cells, telomerase is positive and is expressed. It is convinced that reactivation of telomerase is concerned with tumorigenesis. hTERT acting as determinative factor only is highly expressed in telomerase positive tumor cells, but not in telomerase negative normal cells. hTERT can act as associated antigen of tumor differential expression and is considered to be an ideal target for cancer therapy. This study linked B7.2 gene with hTERT gene and constructed eukaryotic expression vector, building cancer targeting gene therapy system in order to realize cancer targeting therapy. ObjectiveTo construct the eukaryotic expression vector containing both B7.2 and hTERT gene to facilitate further study of cancer targeting gene therapy.
