Macromolecule polymers have many advantages, such as easily machining and low-cost, had been used as the raw materials in the fabrication of microfluidic chips in recent years. Polydimethylsiloxane (PDMS) has the common features of macromolecule polymers, besides that, it had features of highly transparent to the visible lights, fine ability to copy micro- or nano- masters, innocuity, good biological compatibility, and perhaps was the most universal used polymer material in the fabrication of microfluidic chips. The ordinary method to fabrication microfluidic chips by PDMS was casting, that was firstly to fabricate master of the chip, then use PDMS pre-polymer to cast the master, after the curing of PDMS, drill holes and bond them. The fabrication of chip master was the key step, it consisted several steps, including fabrication of photolithographic mask, photolithography, chemical etching. The whole fabrication process was complex and many factors should be considered and controlled carefully. The whole fabrication cycle was long, and the final glass master was very flimsy in its application. The ideal chip fabrication method should be enough simple, fast, flexible and low-cost to face challenges of practical application. This paper elementarily founded a new fabrication method for PDMS chip which had two key processes, Laser ablation to fabricate metal model and dissolved polymethylmethacrylate (PMMA) to cast the model for master fabrication. Besides this, this paper had founded a chip-based capillary electrophoresis and chemiluminescence detection system, and then separated several drugs to estimate performance of the system. The research contents and results of this paper were as following:1) This paper elementarily founded a new fabrication method for PDMS chip which had two key processes, Laser ablation to fabricate metal model and dissolved PMMA to cast the model for master fabrication. This paper confirmed stainless steel as the metal model material, and used CCD microscopically imaging technology to observe and analysis the surficial situation of microchannels, and confirmed preferable Laser ablation parameters. This paper chose dissolved PMMA in chloroform to cast the metal model to form polymer master, and confirmed the preferable proportion of PMMA and chloroform, created a method to use different viscosity solutions to casting models in turn to form fine polymer master. This paper had designed and fabricated a frame for fix master and casting PDMS.
