Objective: To investigate whether tetrandrine combining with celecoxib can reverse multidrug resistance in leukemia cell line K562 and explore its possible mechanism.Methods: Cytotoxic effect of tetrandrine alone and tetrandrine combining with celecoxib on K562 cell line was detected using MTT colorimetry; Apoptosis, intracelular drug concentration, the expression of P-gp and cell cycle were determined by flow cytometry(FCM); The expression of MRP, TOPOâ…¡, GST-Ï€and Cox-2/Cox-1 was detected using RT-PCR.Result: Compared with other groups, combination of tetrandrine and celecoxib has stronger inhibition effect on K562 cell line and higher intracelular drug concentration and apoptisis percentage(17.67%) . Tetrandrine and celecoxib treatment can down regulate the expression of P-gp and arrest the cell cylce at G2 stage.Under such treatment, the mRNA expression of MRP, GST-Ï€ and Cox-2/Cox-1 in K562 cell line decreased, however, TOPOâ…¡ expressin was up-regulated.
