| PART I: Introducing a modified OLT modelObjective: To introduce an animal model of modified cuff technique orthotopic of liver transplantation in ratMethods: 80 cases of rat orthotopic liver transplantation were performed using the two-cuffed technique with some modification in graft harvesting, graft liver perfusing , the anastomoses of suprahepatic vena cava and the perioperative treatment on the basis of Kamada using the cuffed anastomoses of vena . the postoperative syndrome and survival rates were investigated.Results: Of the 40 cases , the mean time of anhepatic period was 15min, and the mean operation time was 45min. The 24 hours and 2 weeks survival rates were 95% and 90% respectively.Conclusions: The modified two-cuffed methods is convenient and easy to duplicated. It can enhance the stability and survival rate of rat orthotopic liver transplantation models.PART II: The protective effect of Ligustrazine on liver graft against IRI in rat OLT modelObjective: To estimated the protection of liver graft against ischemia-reperfusion injury in rat orthotopic liver transplantation by use of Ligustrazine.Methods: 12 couples of SD male rats ,weighed 180-250g, were randomized into experimental group and control group. Perfusing and preserving solution were Ligustrazine Ringer's lactate solution in the experimental group and Ringer's lactate solution without Ligustrazine in the control group. All grafts were preserved for 4 hour then transplanted orthotopically. The bile output was observed in two groups of liver grafts after portal vein reperfusion .Six hours after reperfusion , the recipients were sacrificed , serum ALT,AST,... |
