Objective:To observe the change of cardiac matrix metalloproteinases-3 in the rabbit model of acute myocardial infarction and the relation between MMP-3 and heart function as well as the intervention of Aprovel, to investigate the possible mechanism of Aprovel's function for ventricular remodeling after acute myocardial infarction. Methord: Rabbit myocardial infarction model was established by permanent ligation of left anterior descending coronary artery.68 New Zealand rabbits were randomized divided into 3 groups:sham group,infarcted group and drug treatment group which was divided into 2d,4d,7d,14d(n=3) and 10weeks(n=6).Expression of MMP-3 protein was achieved by immunohistochemical method and Western-Blot. 10 weeks after operation,the interventricular septum thickness(IVSd),left ventricular posterior wall thickness(LVPWd),left ventricular ejection fraction(EF) were measured by echocardiography.Result: There is low level expression of MMP-3 in sham group.In infarcted zone, MMP-3 was increased in infarcted group at 2d,reached the peak at 7d and decreased gradually from 7d . Expression of MMP-3 was higher at 14d compared with 2d. MMP-3 in Aprovel group reached the peak at 4d and decreased gradually from 4d . Expression of MMP-3 was lower at 14d compared with 2d, but was significantly decreased at each subset compared with those of infarcted group(P<0.05).MMP-3 expression of none-infarcted zone has no significant difference within 14 day. At 10th week, IVSd and LVPWd had a significant decrease in drug treatment group compared with those of infracted group(P<0.05).EF was obviously increased (P<0. 05),but still lower than sham group(P<0.05).
