Dendritic cells (DCs) are known as the professional antigen presenting cells (APC), which perform important roles in linking innate and adaptive immunity. Overwhelming evidences demonstrate that DCs are the most potent professional APC. Immature DCs capture antigens at the site of antigen deposition, then migrate to lymphoid organs, where they present the processed antigens to resting T cells and promote the adoptive immune response. Whether DCs can be mature and play their function relates to many factors including antigen stimulation, the expression of costimulatory molecular, and chemokines with their receptors' function. During the process, DCs express costimulatory molecular and secret certain chemokines and switch chemokine receptors' expression according to their.maturation stage. Understand the interaction and internal relationship of these factors play a significant role in designing more effective DC vaccine for clinical use.PBMC derived immature Mo-DCs, which express low-level costimulatory molecule and high-level chemokine receptors CCR1, CCR2, can response to inflammatory chemokine RANTES etc. Immature Mo-DCs have strong migration ability to inflammatory locus, which has high-level inflammatory chemokines in their microenvironment to ingest and dispose antigens. It was reported that tumor constitutions also could merely attract immature Mo-DCs although the chemokines in tumor microenvironment remain unknown. After peripheral immature Mo-DCs ingest antigen and receive other promote-mature factors' stimulations, the gradually mature Mo-DCs excrete plenty of inflammatory chemokines, down-regulate their expression of inflammatory chemokine receptors through autocrine effect, and show digressive response ability to inflammatory chemokines which profit Mo-DCs' migration out of inflammatory...
