Objective: The aim of the research is to investigate the effect on VEGF expression and cell proliferation of human prostate carcinoma cell line PC-3 treated with VEGF antisense oligodoxynucleotide in vitro; meanwhile, this experiment in vivo was to explore a new therapeutic strategy to prostate carcinoma by investigating the effect of VEGF antisense oligodoxynucleotides on VEGF expression, growth and microvessels density of tumor and tumor cell apoptosis.Methods: In vitro: VEGF antisense, sense oligodexynucleotides were added to the medium of cultured PC-3 cells in different concentration with Lipofectin.VEGF expression was detected by FCM technique and PC-3 cell proliferation was assessed by using MTT method. In vivo: PC-3 cells were injected subcutaneously to nude rats,then VEGF antisense, sense oligodexynucleotides and saline were gived into tumors respectively.The volume and weight of tumor were measured.VEGF protein ,microvessels density were detected by immunohistochemical staining and tumor cells apoptosis was assessed by TUNEL method.Results:VEGF expression of PC-3 cells was from 0.037±0.006 to 0.102±0.008 with different concentration antisense oligodeoxymucleotides .PC-3 cells inhibitary rate is raised from 18.55% to 38.99%.VEGF expression and proliferation of PC-3 was inhibited by antisense oligodeoxynucleotides and inhibitory effect is in concentration fashion,however,no inhibitory effect...
