Objective: To investigate the expressions of matrix metalloproteinase 2 (MMP2) , matrix metalloproteinase 9 (MMP9) and E-cadherin and their correlation with infiltration and tumormetastasis in human colorectal cancer, to evaluate the relationship among the three proteins.Methods: Immunohistochhemical staining (streptavidirr-peroxidase method) was used to detect MMP2 , MMP9 and E-cadherin expression in 52 cases of colorectal carcinoma (including 3 cases of colorectal adenomas carcinogenesis), 10 cases of normal colorectal tissues, to analysis the relationship between them and metastasis. The results of existence from 52 cases followed-up had also been analyzed.Results: (1) In normal colorectal mucous, MMP2 and MMP9 were nagetive ,no overexpression was observed . Their expressions in carcinoma were significantly stronger than those in adjacent tissues , no matter in primary cancers or metastatic ones(48/52,47/52) . It is also showed that the over expression of MMP2 and MMP9 in cancer tissues were related to lymph node metastasis status and invasion . the expression of the group with lymph node metastasis(100%,1000%) was higher than that with no lymph node metastasis(91%,89%) . The over expression of them was related to tumor grades , the higher grade (87.%, 83.7%) ,the poorer grade (100%, 95.2%) . Both the over expressions of MMP2 and MMP9 had significant difference between the group of survival time <5 years and that of >5 years (P < 0.05) High expression of E-cadherin was detected in normal colorectal mucous membrane epithelium (100%) while E-cd expression was lower (36.5%) in the tumor samples .
