Objective: To investigate the combined effects of CD40L monoclonal antibody(mAb) with immature dendritic cells on donor-specific immune tolerance in rats undergoing small intestinal transplantation.Methods: 190 SD rats were performed segmental heterotopic small intestinal transplantation with microsurgical technique. And the graft portal vein and aorta including superior mesenteric artery were anastomosed by end to side with recipient inferior vena cava and infrarenal abdominal aorta respectively. The pre-DC was dissociated from donor marrow, and stimulated with GM-CSF and IL-4 in vitro. After five days culture , the expression of co-stimulation molecular and MHC- II was detected. Seven days before transplantation of small intestinal from Wistar donors,2 × 10~6 donor-derived immature DC were infused intravenously into SD recipient rat with or without a concomitant intraperitoneal injection of anti-CD40L mAb. We observed :1.Survival times of recipients after transplantation.2.The pathologic changes of small intestinal and spleen 3,5,7,14days after transplantation.3. Apoptotic death in graft-infiltrating cells and in areas of T-dependent lymphoid tissue was analyzed by terminal deoxynucleotidyltransferase-catalyzed dUTP-digoxigenin nick-end labeling and quantitative spectrofluorometry. 4.Recipient serum IL-2,IL-10,IFN-r concentrations were detected with ELISA kits according to the manafacture's instructions,respectively.Results: A total of 95 small intestinal transplantation were...
