Objective: To investigate the effect of the ICOS antibody on the apoptosis and function of T cell induced islet cells and its mechanisms. Methods: The RINm5f cell and rat T cells were co-cultured with ICOS antibody for 12h. 24h. 48h. (1) The protective effect of ICOS antibody onthe apoptosis of RINm5f cell strain induced by T cells was observed by flow cytometry. (2) At the end of 48h, total cellular RNA was extracted, and the mRNA of the BAX. Bcl-2 and the genes of the apoptotic pathway,such as FAS. TRAIL. Tnfrsfla, TRADD. FADD. Caspase8 were detected by RT-PCR method. (3)The effect of the ICOS antibody on the islet insulin genes INS 1. INS2 and some transcription factors was detected by RT-PCR method.Results: (1) T cell increased the apoptotic rate of islet cell. After co-cultured with ICOS antibody, the apoptotic rate of the RINm5f cells decreased profoundly and y interferon antibody could strengthen the anti-apoptotic effects. (2)The ICOS antibody stimulated the Bcl-2 gene expression and inhibited the BAX gene expression. (3) The ICOS antibody inhibited the gene expression of FAS, TRAIL, Tnfrsfla, TRADD, FADD and Caspase8 of islet cells. (4) Compared with control...
