Object To investigate the interrelationship of Tec tyrosine protein kinase and multidrug resistance induced by induced by DNA damaging chemotherapy. Methods We collected tissues in 35 histologically verified HCC. A single-cell suspension of HCC was prepared and treated with Adriamycin. The cell growth inhibition was evaluated by tetrazolium (MTT) assay. On other hand, total RNA was abstract from the HCC tissues for Slot Blotting to examine the expression of Tec. HepG2 cell were seeded and treated with Adriamycin, Vincristin or Cisplatin. Then the expression of Tec was examined by Western Blot. Tec eukaryotic expression vector were stabile transfected into HepG2 cell. Then the cell was treated with vincristine. The cell survival rate was valuation by typan blue staining. Cell colony formation was detected by Giemsa staining.Results The expressions of Tec were higher in 35 HCC than in the other 13; and chemotherapeutics sensitivity of the 35 HCC was lower than the other 13. Tec protein expression was induced after Vincristin treated. Tec transfected HepG2 cell was more resistant to chemotherapeutics which display increased cell survival rate and enhanced clony formation ability.Conclusions Expression of Tec is positive correlated with drug resistance induced by chemotherapeutics in HCC. Tec might be involved in muti-drug resistance in HCC.
