Objective: To Abstract establish rat model of ocular hypertension, To observe the development Process of retinal injury in ischemia-reperfusion(IR) state, investigate apoptosis occur in retina and study NF-kB expression in a rat retinal ischemia reperfusion and the effect of the panax netoginseng saponins(PNS) on its expression.Methods: 52 Sprague-Dawley rats were devided into three groups at random, containing cornia damaged group, ischemia-reperfusion group(IR), ischemia-reperfusion and PNS group (IR+PNS). 48 rats were subjected to 15kpa and 60minutes of Pressure-induced ischemia, then sacrificed and fixed after ischemia -reperfusion: 2hours,6hours, 12hours, 24hours, 48hours, 72hours, to detected with light microscopy, apoptosis was detected by terminal dUTP nick end labeling (TUNEL).NF-κB was examined in the retina by immunohisto-chemical at light level.Result : Compared with normal control, histopathological changes were revealed in IR group, including retinal edema, vacuolar degeneration, karyopycnosis and karyolysis, derangement of retinal constitution. The longer reperfusion after the ischemia ,the more serious of retinal injury. The histopathologic damage of retina in IR+PNS group was obviously more lighten than in IR group . Tunel positive expression were detected in RGL and IPL of the retina . The highest level of expression occurred 24hours after injury in IR group and its level was higher than the level in the IR+PNS group at every stage (P<0.05).
