Objective: To establish human gemcitabine-resistant non-small cell lung cancer cell lines so as to discuss the influences of inducements to establishments of human gemcitabine-resistant non-small cell lung cancer cell lines, meanwhile, we will invesgate the biological characters of the human gemcitabine-resistant non-small cell lung cancer cell lines and taxol's effect to them. Methods: On the basis of 'high dose intermittent selection' and 'gradually increasing dose selection', The drug-resistant cell lines A549/Gem and H460/Gem were established by clinical serous peak concentration gemcitabine intermittent selection, 2/3 clinical serous peak concentration gemcitabine intermittent selection, repeated clinical serous peak concentration then low but gradually increasing concentration of gemcitabine and 2/3repeated clinical serous peak concentration then low but gradually increasing concentration of gemcitabine from their parental cells human lung adenocarcinoma cell line A549 and human large cell lung cancinoma cell line NCI-H460 who are sensitive to gemcitabine respectively. During the course of inducement, in order to make a preliminary determination of the influences of inducements to establishments of human gemcitabine-resistant non-small cell lung cancer cell lines, we monitored their morphology, resistance index, resistant pedigree, growth curve, doubling time, DNA content et al; at the same time, we have monitored their expression of NSCLC tumor markers, gathered their biological characters from DNA, mRNA and protein levels separatively.
