[Objective] To analyse the 18S rDNA genotype of Acanthamoeba strains isolated from Jilin Yanbian soil (CJY/Sl)and detect the effect of Allitridium on Acanthamoeba spp. [Methods] A strain of Acanthamoeba was isolated from soil and cultured in PYG . Full genome DNA sequences of the strain of Acanthamoeba was amplified using PCR, then the PCR products were sequenced. The 18S rDNA sequence was analyzed by Clustal X software and compared with other strains. By Acanthamoeba castellanii Neff , the influence of Allitridium on the morphological feature, proliferation speed and ultrastructure was tested . [ Results ] The full length of 18S rDNA of the strain CJY/S1 is 2255bp. The sequence similarity is 99% with Acanthamoeba castellanii Neff .The Rns genotype of strain CJY/S1 was T4. At concentrations of 100ul/ml Allitridium forced the Acanthamoeba trophozoites into cysts and inhibited its proliferation speed. The 1000ul/ml groups of Allitridium effectively destroyed the ultrastrucutres of the Acanthamoeba trophozoites.[Conclusions] According to the Rns full genotype classification criterion, the strain CJY/S1 isolated from soil is Rns genotype T4. Therefore, Allitridium is effective in destroying Acanthamoeba.
