Crude drug is gaining increasing popularity worldwide for health promotion and adjuvant therapy. The modernization of crude drug calls for modern scientific methods to study the pharmacological information of active ingredient in molecular level. But in the present research, more attentions are concentrated on the quantitative and identification analysis of the active ingredients or drug standardization. The research of crude drug requires newly advanced thoughts and methods to understand the complexity and compatibility of crude drug. In this thesis, some basic physicochemical constants including hydrolysis rate constant, binding constant and ionization constant of ingredients in crude drug were studied with capillary electrophoresis-electrochemical detection (CE-ED).This thesis consists of five chapters. In chapter 1, the general introduction consist of the current situation of crude drug, the information about the physicochemical constant of crude drug, a brief introduction to CE, and the researches in this thesis were concisely described.In chapter 2, the hydrolysis rate constant and activation energy of main active ingredient in Cortex Fraxini aesculin was studied. Under the optimum conditions, the hydrolysis rate constants of aesculin hydrolysis at five different reaction temperatures were obtained respectively. And the activation energy of the hydrolysis reaction was also calculated.In chapter 3, CE-ED method was developed to make a kinetics study on the hydrolysis reaction of chlorogenic acid. The change of the concentrations of chlorogenic acid and caffeic acid during the process of hydrolysis was directly demonstrated.
