| [Objective]: The enhancement of venule permeability occurs in many diseases, such as wound, burn, shock and inflammation. Especially in burn shock, it plays a key role in the occurrence, development and turnover of this course. Assisted by computer technology, we study the influence of burn to the venule endothelial cells F-actin and tight junction reconstruction, the enhancement of permeability and the effects and mechanism of p38 MAPK in this process. Then elucidate the molecular mechanism of the loss of liquid during burn. We hope to explore new therapy and new drugs and provide new target for the prevention and cure of liquid loss in clinical therapy.[Methods] :By the vascular denudation, we have got the skin venule of normal and burned rats. The skin venules are respectively taken as normal control and experimental group. The venule model is labeled with flurescin and F-actin fluorescence probe coupled with monoclone antibody ZO-1 and is observed by laser scanning copolymerization microscope to compare the distribution and structure of the connexin and F-actin in normal and burned rats. Via computer figures and fluorescence ratio assays, we know the permeability of normal rat skin venule and compared with F-actin and connexin, then discuessed the correlation.The venule model of burned rat is incubated with p38 MAPK specific inhibitor SB2035080 and then label with flurescin and F-actin fluorescence probe coupled with monoclone antibody ZO-1 and is observed by laser scanning copolymerization microscope to find the influence of burn to the distribution, structure and permeability... |
