Objective:To investigate the proliferation and against human ovarian serous carcinoma cell strain activity of NK92 cell induced by interleukin-18 (IL-18) in combination with interleukin-2 (IL-2) in vitro.Method:NK92 cells were induced by IL-18 at different doses in combination with (or) interleukin-2 in vitro. The experiment divide into: IL-2 group, IL-18 group, IL-18 combine with IL-2 group and control group. The proliferation of NK92 cells induced by different concentrations of IL-18 and IL-2 were measured by methyl thiazolyl tetrazolium (MTT) methods, in order to choose the best stimulated concentration. The change of cell cycle analysis was observed by flow cytometry (FCM). Lactate dehydrogenase releasing method (LDHRM) was used to examine NK92 cells mediated cytotoxic activity on the growth of ovarian carcinoma cell lines with different effector-to-target ratios in vitro. TNF-α, IFN-γ levels in supernatant were quantified by enzyme-linked immunosorbent assay (ELISA) when the cytotoxicity was the highest.Results:1.
