Objectives 1. To explore the influence of hypertension on the advanced glycosylation end products (AGEs) which is looked as the signal of aging and it's damage to the structure of the vascular tissue; 2. To explore the change of AGEs receptor (RAGE), NF-κB, and the cytokine such as ERK1, ERK2 etc in the process of hypertensive injury of blood vessel; 3. To evaluate the relative value of Benazepril in reversing the aging and injury of hypertensive vascular. Methods There were 24 spontaneously hypertensive rats (SHR) 30 weeks old were divided into 2 groups randomly: 1.SHR-control group, which were fed distilled water one time per day; 2.SHR- Benazepril group, which were fed Benazepril 10mg/(kg.d); and there were 12 Wistar-Kyoto rats (WKY) which were 30 weeks old were fed distilled water one time per day as the WKY control group. We measured the Systolic blood pressure (SBP) and heart rate of tnem weekly After 12 weeks of treatment, all rats were drawn for detection, including: 1.The level of PRA, AngII(inclouding myocardial AngII level),ET, ALD,NO in blood serum; 2.Vascular pathologic changes were observed. 3.The mRNA expression levels of NF-κB, VCAM-1 were detected by RT-PCR, respectively. 4.The protein levels of blood vessel NF-κB, RAGE, eNOS, iNOS, VCAM-1, ICAM-1, ERK1, ERK2 and ET-1 were detected by the immunohistochemical methods. 5.The level of AGE were measured...
