Anti-apoptotic Actions Of Intranasal Insulin-like Growth Factor Ⅰ And Erythropoietin In The Rats With Epilepsy

Objective: In this study, we investigated the anti-apoptotic action of intranasal Insulin-like growth factorâ… and Erythropoietin in the rats with epilepsy. To make a preliminary assessment about the curative effect intranasal of IGF-1 and EPO. And we also investigated the probalble mechanism of action.Methods: Male SD rats, weighed 200 ~250g. The model of EP (epileptic paroxysm) was set up. 42 severe ones were filted from the rats with EP according to the grade of EP. Then the rats were divided into 4 groups according to the factorial design: Sodium chloride group,IGF-1 group,IGF-1+EPO group,EPO group.The percentage of cell apoptosis was examined by the flow cytomer.The expression levels of caspase-9, XIAP, smac/DIABLO protein were examined by Western Blot.All the results were analyzed on Statistical Package for Social Sciences 13.0.Results: 1. During the process of intranasal administration,the mortality in the 4 groups were not statistically significant(P>0.05)2. The percentage of cell apoptosis was examined by the flow cytometer.EPO guoup can reduce the cell apoptosis better than sham group(P<0.01); IGF-â… group can also reduce the cell apoptosis better than sham group(P<0.01); IGF-â… +EPO group was better than EPO group and IGF-â… group(P<0.01).3. The expression levels of XIAP protein were examined by Western Blot. EPO guoup can improve it's expression better than sham group(P<0.01); IGF-â… group can also improve it's expression better than sham group(P<0.01); IGF-â… +EPO group can do it better than EPO group and IGF-â… group(P<0.05).4. The expression levels of smac/DIABLO protein were examined by Western Blot.
