Effect Of Matrine On HSC Apoptosis In Mouse Acute Liver Injury

AIM To study effects of matrine on HSC apoptosis and NGF secretion of hepatecyte in acute mouse liver injury;approaching the potential mechanism how matrine induces HSC apotosis.Methods 32 KunMing srain clean mouse,22-25g,were separated to 4 group randomly according to mass:control group,matrine 50mg/kg group,matrine 75mg/kg group and matrine 100mg/kg group. CCl4 was administered at 1.6 g of CCl4 per kg by oral gavage,at the same time matrine was injected intramuscularly in mouse lag limb according to different group per 12 house,4 times in all.60 house later animals were sacrificed and their livers harvested,4% formalin-fixed, paraffin-embedded and made to sections. Immunohistochemistry for the expression of NGF and count the grey scale through Image-plus 5.0,double staining ofα-SMA and TUNEL for the HSC apoptosis.Results 1.After treated with matrine at different concentration of 50mg/kg,75mg/kg,100mg/kg for 60 hour,the cross product between the reciprocal of the immunohistochemistry gray scale and the gray areas were 2.969±0.523μm2,5.998±1.021μm2,5.419±0.779μm2 respectively,which are significantly higher than that of control group(0.484±0.106μm2 F=105.433,P<0.01).2. Double staining ofα-SMA and TUNEL shows HSC apoptosis are 2.750±1.282/HP﹑5.625±1.685/HP﹑8.000±2.152/HP,11.375±1.634/HP respectively.The difference is significant;Matrine can significantly promote HSC apotosis in dose-dependent manner(F=37.760,P<0.01), 3.there is positive correlation of the action of matrine between the promotion of NGF expression and the apoptosis of HSC (r= 0.783 ,t=4.17 ,p<0.01).Conclusions matrine can promote HSC apoptosis in acute mouse liver injury,the mechamism may be that It promote NGF expression of hepatacyte and so that promote HSC apotosis through paracrine secretion...
