| Obejective: To observe the relationship among SOD2 expression, Mn and Fe levels, and neurodegeneration induced by chronic administration of aluminum in mice. Methods: 1. Animal model: The neurodegenerative model of mice was established via intragastric administration of AlCl3 solution(element aluminum 400 mg/kg)once a day, 6d a week for 3 months. 2. Learning and memory function test: The step-strough test and morris- water maze were used to assess the changes of learning and memory functions of the experimental mice. 3. Pathomorphology: Histopathological sections of hippocampi were stained routinely with HE and detected under the microscope. 4. Other methods: Inductively coupled plasma atomic emission spectrometry, conventional biochemistry techniques,RT-PCR,and Western-blotting were used to determine the levels of metal ions, MDA contents, activities of SOD and SOD2,as well as expressions of SOD2... |
