The research includes two parts to study the solitary and cooperative effect of retinoic acid (RA) and sonic hedgehog (Shh) on the differentiation of bone marrow stromal cells (BMSCs). In the part one, we cultured the BMSCs in special medium made by our Lab (Patent number: ZL02134314) to master the method of their primary culture and purification, and obverse the condition of proliferation, development of BMSCs. By checking Nestin (a kink of NSCs marker) and Hb9 (an important tag in motor neurons (MN)), we study the biologic character of differentiation in BMSCs. In the part two, by means of classifying four groups (control, RA group, Shh group, RA+Shh group) with the difference of inducing factors, we researched the solitary and cooperative effect of retinoic acid (RA) and sonic hedgehog (Shh) on the differentiation of BMSCs-D-NSCs, and analyzed it upon the regularity of neural development biology, in order to explore the potential of BMSCs-D-NSCs developing into MN.Chapter â… :Research on the culture and biological characters of BMSCs Objective...
