With the monitor of HPLC, two objective compounds were separated with various column chromatography from Paeonia lactiflora Pall, and identified by spectral analysis as: albiflorin and paeoniflorin.A procedure was established in which a composition of two monoterpene glucosides from paeony root was prepared. And it was optimized and used to scale up the preparation of the composition..Quantitative HPLC method was set up to measure albiflorin and paeoniflorin. Several factors were studied systemically such as:linearity, repeatability, stability and recovery, which were accord with conformance to specifications.The percentage of albiflorin and paeoniflorin in composition was determined up to 92%.The contents of albiflorin and paeoniflorin in different parts of Paeonia lactiflora Pall. were determined.The ratio of albiflorin and paeoniflorin were 1:3,1:1 and 4:1 in leaves ,stem and root respectively.Immune activity of albiflorin and paeoniflorin were studied by the tests of macrophage activity, macrophage cytotoxicity and lympholyte multiplication. The results indicated that albiflorin and paeoniflorin could improve activity of macrophage of mice, accelerate lympholyte multiplication,increase amount of active lympholyte,enhance immunity and confront with Cytoxan.
