Garlic is the bulb of Allium satirum L., Garlic oil is the volatile oil extracted by steam distillation or organic solvent. As an antifungal agent clinically, Garlic oil has good curative effects with wide antifungal spectrum and low toxicity. Now Garlic oil injection is withdrawn from markets for its potential hemolytic toxicity caused by Tween-80, a kind of solubilizing agent used to improve the solubility of Garlic oil since Garlic oil is slightly soluble in water. This article chooses submicron emulsion as drug carrier to prepare oil-in-water submicron emulsion of Garlic oil for intravenous administration in order to reduce irritations and toxicities, improve the stability of Garlic oil and provide safe and effective antifungal injection of traditional Chinese herb.GC was established to determine the content of dially trisulfide (DATS) in Garlic oil. Garlic oil was refined by the column chromatography under the decompression. Refinement technique was optimized using orthogonal experimental design.A submicron emulsion was obtained by passing the coarse emulsion through a high pressure homogenizer. The major composition of the formulation was established on the base of classic fat emulsion and signal factor studies. Appling stability parameters of centrifugation, content of DATS and appearances of emulsion to evaluate the physical and chemical stability of emulsions ,the best formulation was established by orthogonal experimental design.
