[Objective] : Compare continuous illumination to intermittent illumination the influence of Wistar mouse retina, discuss retina light damage occur mechanism; After light damage, measure retina GFAP expression, discuss Muller cell the role in retinal light damage.[Methods] : Forty-two 8-10 week-aged, female Wistar mice were randomly divided into normal group, continuous and intermittent damage group.All mice were maintained under cyclic light environment 7 days,and then kept in darkness for 24 hours before the experiment.The green fluorescent lamp of 2380 ±569 Lux was used as injury light source, Continuous damage group were illuminated for 24 hours continuously; Intermittent damage group were illuminated for 3 hours, then put into dark environment for 3 hours,and illuminated for 3 hours again,repeat 8 times. After illumination, 3, 7 and 14 days take eyeball, paraffin bale is buried , HE dyeing and Immunohistochemical staining of GFAP, Image analyzer measure retina ONL layer thickness and GFAP gray scale worth. With SPSS software package, make statistics of analysis.[Results] : Normal Wistar mouse retina has distinct structure normally,IS and OS have distinct boundary, INL and ONL put in order closely, ONL thickness is 43.49± 1.45μm; After 24 hours of illumination, Every damage group has obvious pathological change , show mainly is: IS and OS structure are disorder ,their boundary is not clear, ONL nucleus gap increases , OPL and ONL change thin.
