Objective: In order to isolate, purify and proliferate rabbit bone marrow-derived mesenchymal stem cells(MSCs). To investigate the migration and differentiation of transplanted autologous MSCs in myocardium after the acute myocardial infarction and the feasibility of treatment of myocardial infarction by transplantation of autologous MSCs.Method: Bone marrow was density gradient centrifuged and cultured in glass culture bottle to isolate and proliferate MSCs. The growth curve was drawn. The cells were examined by invert microscope and transmission electron microscope. 20 New Zealand white rabbits were divided randomly into two groups: acute myocardial infarction control group (AMI group, n=5, the descending anterior branch of left coronary artery was ligated and PBS was injected into the periinfarcted tissues 1 hour after the ligation); acute myocardial infarction + autologous MSCs transplantation group (AMI + MSCs group, n=15, 1 hours after the ligation , DAPI labeled MSCs were injected into the periinfarcted tissues). 10 minutes 3 days and four weeks after the implantation, the rabbits were sacrificed and their hearts were harvested.
