The Study On Pharmacokinetics Of Shuluo Effective Compounds

Traditonal Chinese Medicines and decoction were characteristic with many effective compounds, diversity of active constituentsand pharmacological mechanism,and comprehension effection using in clinic widely. The studies were focus on studing pharaiacokinetics of Chinese herbal combination So. The effective compounds of Shuluo on cerebral ischemia reperfusion were studied in this papers.This dissertation consists of there parts: pharmacokinetics in rats, the penetrativity of blood brain barrier in MCAO rats, and relativity between penetrativity of blood brain barrier and P-glycoprotein 1. Pharmacokinetic and tissue distribution of ShuluoObjective to investigate pharmacokinetic parameters and tissue distribution performance of Geniposide and paeoniflorin(PF) contained in Chinese recipe Shuluo in rats. Methods Blood and tissue samples were collected before and at various time after injecting vein tow doses of soluo injection in rats.Then, prepared the sample on demand and determined them by RP-HPLC. The concerntration-time curves were analyzed with 3P87 program to determine the compartment models and pharmacokinetic parameters; Results (1) The conditions of chromatography: The column was YWG-C₁₈(250mm×4.6mm,10μm),The mobile phase for Blood and tissue samples was acetonitrile:PBS:water: (15:5:85).The Uv detector was set at 240nm for Geniposide and 232nm for paeoniflorin(PF) respectively.The flow rate was 1.0 ml/min, temperature is maintained at 30℃.
