Cholera is an acute diarrheal disease with characteristics of spreading quickly and severe damage. It is the first class of quarantined diseases in China and one of world seven pandemic diseases. There had been seven extensive epidemics in the world since 1817. Cholera enterotoxin (CT) composed of one A subunit (CTA) and five B subunits (CTB) is an major causative agent of vibrio cholerae. As a better immunogen and mucosa immunoadjuvant it can be use to develop mucosa vaccine. At present research of mucosa vaccine on CTB subunit proteion and ctxB gene of cholera toxin is more, but that of CTA subunit proteion and CT is less, in addition, there are not any reports about simultaneous comparison adjuvant activities of ctxA, ctxB and fusion ctxAB genes in vivo, and the mechanism of immunoadjuvant activity about CT isn't completely clear yet, so its application is not widespread to some extent. Therefore, this study choose the subject of cholera enterotoxin gene (ctx) as research object, and firstly combine ctxA with ctxB by overlap PCR, and construct three kinds of molecular adjuvant DNA vaccine, ctxA,ctxB and ctxAB.
