Objective: 1.To study the effects of long-term allopurinol treatment on cardiac function,ventricular remodeling and proinflammatory cytokines in rabbits after myocardial infarction.2.To investigate the mechanism of the benefit effects of allopurinol on cardiac function and ventricular remodeling after myocardial infarction. Methods: 1.Experimental objective:New Zealand rabbits,male,2.0 to 2.5kg body weight.2.Experimental groups: 18 rabbits were randomly devided into three groups(n=6).Group I was sham-operated group,group II was control group and group III was allopurinol-treated group.Sham-operated group was only opened thoracis and pericardial cavity,control group and allopurinol-treated group were myocardial infarction group.Myocardial infarction was produced by myocardial cryoinjury created by liquid nitrogen.All rabbits began to take foods on the first day after operation,allopurinol(40mg/kg) was administered daily to the rabbits of group III and the other rabbits were given placebo.The body weight was measured every week to adjust the daily dose of drug or placebo.3.Experimental protocols:(1)Left ventricular end-diastolic diameter (LVEDD) and left ventricular end-systolic diameter (LVESD) were measured by echocardiography at the 4 weeks after operation,and then left ventricular ejection (LVEF) was counted.(2)Haemo- dynamics parameters such as left ventricular end-systolic pressure (LVESP), left ventricular end-diastolic pressure (LVEDP), maximum LV pressure rising rates (+dp/dtmax) and maximum LV pressure decreasing rates (— dp/dtmax) were recorded with a...
