Extensive skin loss from a variety of conditions is associated with significant functional morbidity and even loss of life. Nowadays extensive deep burn wounds is mainly treated by methods of transplatation with split thickness skin autograft. However in many cases, especially extensive burns and other severe skin loss diseases, early and effective closure on wounds is prevented by a limited number of donor sites available for skin grafts. Some methods now widely being used in clinic, always lack of dermis, such as small stamps of autografting, mesh skin grafting, intermingled transplantation of auto-allografting, micro-skin-grafting and cultured epidermal Autog -rafting, can't resolve the problems regarding the cosmesis and function satisfactorily. However, grafting thicker skin may cause donor sites morbidity. Being in the clinic dilemma, there have been constructed a variety of skin substitutes by methods of tissu-engineering in recent years, but their clinical effects are not confirmed and satisfactory. Therefore, a permanent skin substitute with ideal functions will be of clinical importance. Objective: To investigate the feasibility of constructing composite skin by methods of tissue-engineering. To construct tissue-engineered skin with cultured epidermal stem cells and acellular dermal matrix and to investigate the feasibility of repairing animal skin defection with the tissue-engineered skin .Methods : Epidermis was separated from dermis of child's forskin and dissociated into single cells. 1×10~6/ml epidermal cells were...
