Background Hepatic stem cells (HSC) are very important in both theoretical researches and clinical application.On the one hand, hepatic stem cells may participate in the liver regeneration and restoration during serious liver injury. Therefore, the researches on their proliferation, diferentiation and the mechanisms that regulate the proliferation and diferentiation of hepatic stem cells are helpful to elucidate the mechanism of liver development. On the other hand, hepatic stem cells are able to proliferate in vitro culture, and can be induced to diferentiate into hepatocytes, which will provide a new approach of cell transplantation, tissue engineering and gene therapy for liver diseases.The question whether hepatic stem cells exist in livers has been debated for a long time. Due to the lack of specific surface markers, it is rather dificult to isolate and identify hepatic stem cells.There are few hepatic stem cells in adult livers, and these cells normally remain dormant. It is thus necessary to establish an excellent model of activation in order to obtain suficient hepatic stem cells with high viability.Objective To establish hepatic oval cells proliferation model and investigate the isolation, purification and identification of hepatic ovel cells in vitro.
