[Objective]Stat-3(The signal transducer and activator of transcription-3 )participates in cell growth, differentiation, proliferation and subsistence. It was reported that Stat-3 expressed unconventionally in different type of malignant tumors. It is reported: the antitumor immunologic function increased, it might have relationship with the activity lost of immunosuppressive factor of tumor, and the decreasing direct inhibition of tumor to host. The developing of research, especially the research on mechanism of signal transduction brings a new idea to the research of tumor immunologic inhibition mechanism. It has been found that in most human tumor cells Stat-3 signal transduction pathway is unconventionally activated and that Stat-3 that unconventionally activated has relationship not only with tumor cells occurrence and survival, but also with the factor that has function to inhibited immune system secreted by tumor cells. Which is to say, if we can approve Stat-3 signal pathway is not only necessary for tumor cells occurrence and survival, but also has function in tumor immunologic inhibition, we can know that Stat-3 signal pathway not only can cause tumor cells death, but also possibly release or partly reverse tumor immunologic inhibition, then remove body strong immune system participate in killing tumor cells. This research is mostly detect the expression of immunosuppressive factor of human being hepatic cancer cell Smmc7721's Stat3mRNA and VEGFmRNA and TGF-βmRNA. By using JAK kinase inhibitor AG490 on interdicting Stat-3 signal transduction pathway, and comparing of the...
