Polyacrylamide Hydrogel, which is widely used in ophthalmic operations, drug treatment, food packaging products, and water purification etc, is a watery gel consisting of approximately 2.5% cross-linked polyacrylamide and nonpyrogenic water. Recently, Polyacrylamide Hydrogel (PAMG) has been applied in the sector of cosmetic medicine. Since 1997, the hydrophilic polyacrylamide gel, which is imported from Ukraine, has been used for augmentation mammaplasty in China. But its safety is still in doubt in many countries. In the product, there is inevitably residual acrylamide monomer, which is said to have neurotoxicity, reproductive toxicity, genotoxity, developmental and potential carcinogenic effects. Moreover, the combine effect of the remaining initiator, cross-linking agent and AM is also worth exploring.In our research, we firstly presented an approach to determine the amount of residual AM in the PAMG. An ODS-3 column was used to detect the concentration of AM in the extract of PAMG, and the mobile phase was 0.9% NaCl. It showed that this is a practical and simple method for determining AM in PAMG The limit of detection for AM can reach to ppb level, so it can fully meet the requirement of this study.We analyzed the cell biological reaction with human fibroblast, which is rightly one of the actual cell types in human tissue environment, by cell counting, MTT assay,...
