Aim: This study aimed at the biological effect of human platelet supernatant (hPS) on therabbit growth-plate chondrocytes cultured in monolayer and incubated in pellet culture, and to evaluate whether it is an optional source for cartilage tissue engineering.Methods: (l)Growth-plate chondrocytes were isolated from rabbit costal cartilage andcultured in monolayer. hPS was added in medium at different concentrations. Cells growth was detected by WST-8; the production of proteoglycans and the total contents collagen was determined by Alcian Blue method and the hydroxyproline content assay, respectively. ALP activity was measured by modified enzyme dynamical method. (2) Growth-plate chondrocytes were incubated in pellet culture. hPS was added into medium according to the result of previous research. The histological structure of neocartilage was observed by species stained with H&E; The DNA content of neocartilage was measured by hoechst33258 dye; the production of matrix was estimated from the determination of hydroxyproline content and Alcian Blue method. The expression of collagen I and collagen II were detected by Immunohistochemical analysis and RT-PCR determination.Result: (1)In monolayer system: supplement of hPS activated chondrocytes proliferation and induced marked synthesis of GAG, whereas had no effect on the cell content of total...
