Alzheim's disease(AD) is an aging related neurodegenerative disease. Since AD severely influences the health and living quality of elderly people, and brings much of burdens to family and society, its research becomes the focus of aging disease, significantly on the study of pathological mechanism , therapeutic drug and vaccine about AD .The accumulation of Aβ(β-amyloid peptide) is the primary influence driving AD pathogenesis. β-secretase(BACE), is the key cleaving enzyme which plays a very important role in APP(β-site amyloid precursor protein) processing that participates in the secretion of Aβ, so BACE is regarded as a primary target for therapeutic drug design. It should go through the evaluation of safety and validity when AD's new drugs or vaccines targeting to BACE are developed and actually the most important part of validity assessment is to construct cellular model. Owing to the reasons above, construction of the cellular models for BACE research will have great significance. Moreover, due to the important role that the study of BACE take in the therapy or clinical treatment of AD, and so far most of the researching work using neuron cell lines as the models which have difficulties in culturing , and the transgenic animal models used for AD study also have some problems such as expensive price and can not keep the stability of genotype during reproduction, we hope that we could receive the same study goal using 2BS cell line which is more easy to culture, that is to say if BACE has the same expression status and higher activity as the neuron cell model...
